Mechanisms of resistance to anthracyclines and anthraquinones in human breast cancer cells.
The mechanisms of resistance of human breast cancer cell lines MCF-7/A1 and MCF-7/ADR to mitoxantrone, doxorubicin, and daunomycin were examined. The sensitivity of the MCF-7/A1 cells to the anthracyclines was decreased by the presence of p-glycoprotein, the product of the MDR1 gene, as determined by flow cytometry. In addition, the presence of the p-glycoprotein was confirmed by immunohistochemical staining with the use of an anti-p-glycoprotein monoclonal antibody. However, the presence of the p-glycoprotein did not account for resistance to anthracyclines in MCF-7/A1 cells as indicated by the results of exposure to inhibitors of the p-glycoprotein transporter. MCF-7/ADR cells were resistant to daunomycin and had an increased ability to transport the drug into the cell. The relative contribution of this transport mechanism to the resistance of MCF-7/ADR cells was investigated by examining the binding of 3H-daunomycin to membranes and its efflux by determining the accumulation of the drug in cells treated with the drug. The 3H-daunomycin binding to membranes was decreased in MCF-7/ADR cells compared with parental MCF-7 cells, but the difference in 3H-daunomycin accumulation between MCF-7 and MCF-7/ADR cells was not statistically significant. This suggests that transport of daunomycin into the cell is not the sole mechanism of resistance. A possible mechanism of resistance is that the drug is concentrated in the cell membranes, and that this resistance may be overcome by increased drug efflux from the cell.The current invention 01e38acffe
